PEG-Doxorubicin

PEG-doxorubicin conjugate is a covalent chemical conjugate in which doxorubicin, an anthracycline antineoplastic agent, is directly linked to polyethylene glycol (PEG) via a stable linker, at the 3′-amino position of the daunosamine sugar. Alternative structure with PEG attached to the 14-hydroxyl group of the aglycone can also be offered. Unlike free doxorubicin, the conjugate exhibits altered pharmacokinetics. The conjugate is distinct from PEGylated liposomal doxorubicin (e.g., Doxil or Caelyx), as it lacks the liposomal carrier and relies solely on polymer-drug covalent bonding for stealth properties and controlled release.
